RESUMO

Nicotianagandarela Augsten & Stehmann (Solanaceae), sp. nov., a small 'tobacco' known only from one locality at Serra do Gandarela, in the state of Minas Gerais, Brazil, is described and illustrated. It is morphologically characterized by its rosulate basal leaves, red corolla with a short tube not inflated at the apex, and the peculiar habitat, a shaded site under a rocky outcrop ledge along a forested stream. Phylogenetic analyses based on a combined dataset of nuclear (ITS) and plastid (ndhF, trnLF, and trnSG) DNA sequences revealed that the species belongs to the Nicotianasect.Alatae and is sister to the clade with the remaining species in the section. A key for the identification of Brazilian species of the section is given. The unusual habitat, the small population size, and the intense pressure of mining activities in the surroundings made the species assessed as Critically Endangered (CR), needing conservation efforts to avoid its extinction.

PREMISE: Evolutionary studies require solid phylogenetic frameworks, but increased volumes of phylogenomic data have revealed incongruent topologies among gene trees in many organisms both between and within genomes. Some of these incongruences indicate polytomies that may remain impossible to resolve. Here we investigate the degree of gene-tree discordance in Solanum, one of the largest flowering plant genera that includes the cultivated potato, tomato, and eggplant, as well as 24 minor crop plants. METHODS: A densely sampled species-level phylogeny of Solanum is built using unpublished and publicly available Sanger sequences comprising 60% of all accepted species (742 spp.) and nine regions (ITS, waxy, and seven plastid markers). The robustness of this topology is tested by examining a full plastome dataset with 140 species and a nuclear target-capture dataset with 39 species of Solanum (Angiosperms353 probe set). RESULTS: While the taxonomic framework of Solanum remained stable, gene tree conflicts and discordance between phylogenetic trees generated from the target-capture and plastome datasets were observed. The latter correspond to regions with short internodal branches, and network analysis and polytomy tests suggest the backbone is composed of three polytomies found at different evolutionary depths. The strongest area of discordance, near the crown node of Solanum, could potentially represent a hard polytomy. CONCLUSIONS: We argue that incomplete lineage sorting due to rapid diversification is the most likely cause for these polytomies, and that embracing the uncertainty that underlies them is crucial to understand the evolution of large and rapidly radiating lineages.

Abstract speciesLink is a large-scale biodiversity information portal that exists thanks to a broad collaborative network of people and institutions. CRIA's involvement with the scientific community of Brazil and other countries is responsible for the significant results achieved, currently reaching more than 15 million primary biodiversity data records, 95% of which are associated with preserved specimens and about 25% with high-quality digital images. The network provides data on over 200,000 species, of which over 110,000 occur in Brazil. This article describes thematic networks within speciesLink, as well as some of the most useful tools developed. The importance and contributions of speciesLink are outlined, as are concerns about securing stable budgetary support for such biodiversity data e-infrastructures. Here we review the value of speciesLink as a major source of biodiversity information for research, education, informed decision-making, policy development, and bioeconomy.


Resumo speciesLink é um portal de informações em larga escala sobre biodiversidade, que existe graças a uma ampla rede colaborativa de pessoas e instituições. O envolvimento do CRIA com a comunidade científica do Brasil e de outros países é responsável pelos resultados expressivos alcançados, atingindo atualmente mais de 15 milhões de registros de dados primários de biodiversidade, sendo 95% associados a espécimes preservados e cerca de 25% a imagens digitais de alta qualidade. A rede fornece dados sobre mais de 200.000 espécies, das quais mais de 110.000 ocorrem no Brasil. Este artigo descreve as redes temáticas do speciesLink, bem como algumas das ferramentas mais úteis desenvolvidas. A importância e as contribuições do speciesLink são destacadas, assim como as preocupações em garantir um apoio financeiro estável para e-infraestruturas de dados sobre biodiversidade. Aqui revisamos o valor do speciesLink como uma das principais fontes de informação sobre biodiversidade para pesquisa, educação, tomada de decisão, desenvolvimento de políticas e bioeconomia.

RESUMO

Two new species of Athenaea Sendtn. (Solanaceae) from the Brazilian Atlantic Forest are described and illustrated. Athenaea altoserranae I.M.C. Rodrigues & Stehmann, sp. nov. from the Serra do Mar range, in São Paulo State and Athenaea hunzikeriana I.M.C. Rodrigues & Stehmann, sp. nov. from a restricted area in the north-eastern region of Minas Gerais State and the southern part of Bahia State. Both species have brown to purple-brown or purple mature fruits, a character not found in other species of Athenaea. Descriptions, illustrations, complete specimen citations and maps of both species are provided. A dichotomous key to all species of Athenaea is also presented.


ResumoDuas espécies novas de Athenaea Sendtn. (Solanaceae) da floresta atlântica brasileira são descritas e ilustradas. Athenaea altoserranae I.M.C. Rodrigues & Stehmann, sp.nov. conhecida apenas da floresta atlântica da Serra do Mar, no estado de São Paulo, e Athenaea hunzikeriana I.M.C. Rodrigues & Stehmann, sp.nov., conhecida somente de uma área restrita na região nordeste do estado de Minas Gerais e sul da Bahia, divisa entre os estados. Ambas as espécies apresentam frutos amarronzados a arroxeados, um caractere até então não observado em outras espécies de Athenaea. São disponibilizadas descrições, ilustrações, citações completas de espécimes e um mapa de distribuição. Uma chave dicotômica de identificação para todas as espécies de Athenaea também é apresentada.

ETHNOPHARMACOLOGICAL RELEVANCE: species of Terminalia (Combretaceae) are used to treat diabetes and metabolic disorders in Asia, Africa, and America. Terminalia phaeocarpa Eichler is an endemic tree from Brazil, popularly known as capitão. This species is closely related to Terminalia argentea Mart., also vulgarly known as capitão, a native but not endemic tree. Due to their phenotype similarity, these species might eventually prove inseparable and they are indistinctly used by locals to treat diabetes, among other diseases. The potential antidiabetic effect of T. argentea has been previously reported, whereas the biological effects and chemical composition of T. phaeocarpa have never been addressed so far. AIM OF THE STUDY: investigate the hypoglycaemic effect of an ethanol extract (EE) of T. phaeocarpa leaves and its ethyl acetate (FrEtOAc) and hydromethanolic (FrMEOH) fractions, in addition to their activity on the release of pro-inflammatory mediators and inhibition of lipase, α-amylase, and α-glucosidase enzymes. Additionally, it aimed to characterize the chemical composition of the extract and fractions, seeking to identify the compounds related to the biological activities. MATERIALS AND METHODS: The effect on the release of TNF-α, IL-1ß, and CCL-2 was evaluated in LPS-stimulated THP-1 cells (ATCC TIB-202). The inhibition of lipase, α-amylase, and α-glucosidase was tested in vitro, whereas the hypoglycemic effect was assayed in the oral starch tolerance test. The chemical composition was investigated by extensive UHPLC-DAD-ESI-MS/MS analyses. RESULTS: The extract and derived fractions reduced TNF-α (EE pIC50 = 4.58 ± 0.01; FrEtOAc pIC50 = 4.69 ± 0.01; FrMeOH pIC50 = 4.54 ± 0.02) and IL-1ß (EE pIC50 = 4.86 ± 0.02; FrEtOAc pIC50 = 4.86 ± 0.02; FrMeOH pIC50 = 4.75 ± 0.01) release by LPS-stimulated THP-1 cells in a concentration-dependent manner, whereas the inhibitory effect on CCL-2 release did not reach a clear linear relationship for the tested concentrations. The extract and fractions also inhibited in vitro the activity of lipase (EE pIC50 = 3.97 ± 0.12; FrEtOAc pIC50 = 3.87 ± 0.04; FrMeOH pIC50 = 3.67 ± 0.14), α-amylase (EE pIC50 = 4.46 ± 0.27; FrEtOAc pIC50 = 5.47 ± 0.27; FrMeOH pIC50 = 4.26 ± 0.22), and α-glucosidase (EE pIC50 = 5.46 ± 0.05; FrEtOAc pIC50 = 5.79 ± 0.11; FrMeOH pIC50 = 5.74 ± 0.05). The pIC50 values of the test samples were lower than those obtained with orlistat (7.59 ± 0.08) and acarbose (6.04 ± 0.37 and 7.63 ± 0.04) employed as the positive controls respectively in the lipase, α-amylase, and α-glucosidase assays. When assayed in the oral starch tolerance test, the extract and fractions also reduced animal glycaemia. UHPLC-DAD-ESI-MS/MS analyses of the extract and fractions led to the identification of 38 phenolic compounds, mainly phenolic acids, ellagitannins and flavonoids, among others, all of them first-time described for the species. CONCLUSION: Based on our findings, T. phaeocarpa has hypoglycaemic activity and polyphenols are the probable bioactive compounds, which support the ethnomedical use of the species.

RESUMO

The hexane and ethanol extracts from Himatanthus bracteatus (Apocynaceae) stems were evaluated for antiviral activity against Zika virus, yellow fever virus and dengue virus 2 and for cytotoxicity in Vero cells by MTT assay. The ethanol extract showed good antiviral activity against the three viruses with selective indexes (SI) > 10 and its fractionation led to the isolation of the known plumieride that was active only against Zika virus (SI of 15.97).

The Solanum glaucophyllum Desf. has been used to treat and prevent diseases in human and veterinary medicine. On the other hand, plant poisoning causes several bone diseases, among them osteoporosis, which is characterized by osteoblastic hypoplasia. Because the osteoblast is a cell derived from the differentiation of mesenchymal stem cells (MSCs) from bone marrow, the hypothesis is that the plant reduces the osteogenic differentiation of MSCs. The objective of this study was to evaluate the effects of S. glaucophyllum Desf. extract on MSCs cultured in osteogenic differentiation medium. We determined by liquid chromatography that 1 ml of plant extract contained 3.8 µl of 1,25(OH)2 D3 (calcitriol). Four groups of MSCs cultivated in osteogenic medium were evaluated as follows: (a) treated with 100 µl of extract/L containing 0.4 µg/L of calcitriol; (b) treated with 1 ml of extract/L containing 4 µg/L of calcitriol; (c) treated with 5 ml of extract/L containing 20 µg/L of calcitriol; and (d) a control group without extract. We performed alkaline phosphatase activity assay, analysis of MTT conversion to formazan, and evaluated the percentage of cells, and number and diameter of mineralization nodules. The expression of gene transcripts for osteopontin, bone sialoprotein and BMP-2 was analysed by RT-qPCR. After 21 days, there was a significant reduction in MTT conversion to formazan in treated groups, of the cellularity in the group with 5 ml of extract/L, and in the number and size of mineralization nodules in the groups treated with 1 and 5 ml of extract/L. The 5 ml extract/L concentration also reduced transcript expression of osteopontin. It is concluded that S. glaucophyllum Desf. at concentrations of 1 and 5 ml extract/L reduced mineralized matrix synthesis in MSCs cultivated in osteogenic differentiation medium, which suggests that this is one of the mechanisms by which osteoporosis occurs in intoxicated animals.

Capsicum carassense Barboza & Bianchetti sp. nov., a species from mid-elevation of the Atlantic Forest (Minas Gerais, Brazil) is described and illustrated. This endemic new species is distinguished from the most similar C. mirabile Mart. by its moderate to dense general pubescence, narrowly elliptic leaves and larger calyx appendages and corollas. A key for the native Brazilian species of Capsicum growing in the state of Minas Gerais is also provided.

Solanum hydroides Gouvêa & Giacomin, sp. nov., is described from central Brazilian Atlantic Forest. It is known from only three localities in Espírito Santo and Minas Gerais states, where granitic/gneissic outcrops (inselbergs or sugar loaves) are ubiquitous. The new species, here described, belongs to Solanum subgenus Leptostemonum (or the Leptostemonum clade; i.e. the spiny solanums) and is morphologically related to S. hexandrum Vell. and S. sublentum Hiern, with which it shares the shrubby habit, decurrent leaf bases and well-developed calyces that become accrescent, covering glabrous fruits. Solanum hydroides is unique in its combination of comparatively more delicate habit, indumentum of exclusively stellate eglandular trichomes, accrescent but never inflated fruiting calyces that only partially cover the fruits and comparatively shortly lobed and strictly white corollas. The species is threatened with extinction and assessed as Vulnerable (VU), based on the IUCN criteria.

Ela é conhecida apenas para três localidades, nos estados do Espírito Santo e Minas Gerais, em uma região onde afloramentos graníticos/gnáissicos (inselbergues ou pães de açúcar) são onipresentes. A nova espécie pertence a Solanum subgen. Leptostemonum (ou Solanum clado Leptostemonum, i.e. espécies espinhentas com tricomas estrelados) e é morfologicamente semelhante a S. hexandrum Vell. e S. sublentum Hiern, com quem compartilha o hábito arbustivo, a base das lâminas foliares decurrentes e cálices bem desenvolvidos, acrescentes em seus frutos glabros. Solanum hydroides é, contudo, a única espécie que apresenta a seguinte combinação de caracteres: hábito delicado, indumento composto apenas por tricomas estrelados egandulares, cálices frutíferos acrescentes, mas nunca inflados, que cobrem parcialmente os frutos e corolas com lóbulos curtos e estritamente brancas. A espécie é considerada como ameaçada de extinção, na categoria Vulnerável (VU), segundo os critérios da IUCN.

Traditional conservation techniques for mapping highly biodiverse areas assume there to be satisfactory knowledge about the geographic distribution of biodiversity. There are, however, large gaps in biological sampling and hence knowledge shortfalls. This problem is even more pronounced in the tropics. Indeed, the use of only a few taxonomic groups or environmental surrogates for modelling biodiversity is not viable in mega-diverse countries, such as Brazil. To overcome these limitations, we developed a comprehensive spatial model that includes phylogenetic information and other several biodiversity dimensions aimed at mapping areas with high relevance for biodiversity conservation. Our model applies a genetic algorithm tool for identifying the smallest possible region within a unique biota that contains the most number of species and phylogenetic diversity, as well as the highest endemicity and phylogenetic endemism. The model successfully pinpoints small highly biodiverse areas alongside regions with knowledge shortfalls where further sampling should be conducted. Our results suggest that conservation strategies should consider several taxonomic groups, the multiple dimensions of biodiversity, and associated sampling uncertainties.

Recent debates on the number of plant species in the vast lowland rain forests of the Amazon have been based largely on model estimates, neglecting published checklists based on verified voucher data. Here we collate taxonomically verified checklists to present a list of seed plant species from lowland Amazon rain forests. Our list comprises 14,003 species, of which 6,727 are trees. These figures are similar to estimates derived from nonparametric ecological models, but they contrast strongly with predictions of much higher tree diversity derived from parametric models. Based on the known proportion of tree species in neotropical lowland rain forest communities as measured in complete plot censuses, and on overall estimates of seed plant diversity in Brazil and in the neotropics in general, it is more likely that tree diversity in the Amazon is closer to the lower estimates derived from nonparametric models. Much remains unknown about Amazonian plant diversity, but this taxonomically verified dataset provides a valid starting point for macroecological and evolutionary studies aimed at understanding the origin, evolution, and ecology of the exceptional biodiversity of Amazonian forests.

Although Brazil is a megadiverse country and thus a conservation priority, no study has yet quantified conservation gaps in the Brazilian protected areas (PAs) using extensive empirical data. Here, we evaluate the degree of biodiversity protection and knowledge within all the Brazilian PAs through a gap analysis of vertebrate, arthropod and angiosperm occurrences and phylogenetic data. Our results show that the knowledge on biodiversity in most Brazilian PAs remain scant as 71% of PAs have less than 0.01 species records per km2. Almost 55% of Brazilian species and about 40% of evolutionary lineages are not found in PAs, while most species have less than 30% of their geographic distribution within PAs. Moreover, the current PA network fails to protect the majority of endemic species. Most importantly, these results are similar for all taxonomic groups analysed here. The methods and results of our countrywide assessment are suggested to help design further inventories in order to map and secure the key biodiversity of the Brazilian PAs. In addition, our study illustrates the most common biodiversity knowledge shortfalls in the tropics.

RESUMO

A new species of Solanum (Solanaceae) from the Geminata clade is described for the Brazilian flora. Solanum lagoense Stehmann is only known from Lapinha, a rocky massif located in the Lagoa Santa karst region of Minas Gerais State. The flora of this area, including Solanaceae, was studied in detail in the second half of the 19(th) century by the Danish botanist Eugene Warming. The species differs from other members of the Geminata clade in Brazil in its geminate leaves of different sizes, simple multicellular trichomes present on the new growth and young stems, short extra-axillary inflorescences with few (1-3) flowers, and its stellate corollas with cucullate and strongly reflexed lobes. Here we present a description, taxonomic comments and a preliminary assessment of conservation status of this critically endangered species.


ResumoUma nova espécie de Solanum (Solanaceae) pertencente ao clado Geminata é descrita para a Flora do Brasil. Solanum lagoense Stehmann habita o sub-bosque da Floresta Estacional associada a afloramentos calcários junto à gruta da Lapinha, em Lagoa Santa, Minas Gerais, uma área inventariada em detalhe por Eugene Warming, no século XIX. A espécie é distinta das demais Geminata, por apresentar folhas geminadas de tamanhos desiguais, ramos apicais com tricomas simples, multicelulares, inflorescência extra-axilar, curta, com poucas flores (1-3), corola estrelada, reflexa e cuculada. São apresentadas a descrição da espécie, comentários taxonômicos, bem como a avaliação que sugere o status de Criticamente Ameaçada à espécie.

A new section of Eugenia (Myrtaceae) is described, segregate from Eugenia sect. Phyllocalyx. Phylogenetic studies suggest that Eugenia sect. Phyllocalyx as traditionally delimited is paraphyletic. To maintain the monophyly of each of the sections in Eugenia s.l., we herein opt to circumscribe a new section and recognize six taxa in sect. Speciosae, which has a distribution mostly in southeastern Brazil and northern South America. Nomenclatural notes are made and a taxonomic key is provided for the species of the section.

Knowledge of the geographic distribution of plants is essential to underpin the understanding of global biodiversity patterns. Vascular epiphytes are important components of diversity and functionality of Neotropical forests but, unlike their terrestrial counterparts, they are under-represented in large-scale diversity and biogeographic analyses. This is the case for the Atlantic Forest - one of the most diverse and threatened biomes worldwide. We provide the first comprehensive species list of Atlantic Forest vascular epiphytes; their endemism patterns and threatened species occurrence have also been analyzed. A list with 2,256 species of (hemi-)epiphytes - distributed in 240 genera and 33 families - is presented based on the updated Brazilian Flora Checklist. This represents more than 15% of the total vascular plant richness in the Atlantic Forest. Moreover, 256 species are included on the Brazilian Red List. More than 93% of the overall richness is concentrated in ten families, with 73% represented by Orchidaceae and Bromeliaceae species alone. A total of 78% of epiphytic species are endemic to the Atlantic Forest, in contrast to overall vascular plant endemism in this biome estimated at 57%. Among the non-endemics, 13% of epiphytic species also occur either in the Amazon or in the Cerrado - the other two largest biomes of Brazil - and only 8% are found in two or more Brazilian biomes. This pattern of endemism, in addition to available dated phylogenies of some genera, indicate the dominance of recent radiations of epiphytic groups in the Atlantic Forest, showing that the majority of divergences dating from the Pliocene onwards are similar to those that were recently reported for other Neotropical plants.

Two additions and four new species are described from Brazil for the large Geminata clade (Solanum: Solanaceae) bringing the total diversity in the group to 149 species, with 44 of these occurring in Brazil. New species are described from Brazil: Solanumamorimii S.Knapp & Giacomin, sp. nov. from Bahia and adjacent Minas Gerais states, Solanumfilirhachis Giacomin & Stehmann, sp. nov. from Espirito Santo, Solanumpsilophyllum Stehmann & Giacomin, sp. nov. from Minas Gerais and Solanumverticillatum S.Knapp & Stehmann, sp. nov. from São Paulo, Rio de Janeiro and Minas Gerais. Modern character-rich descriptions and lectotypifications are provided for Solanumapiahyense Witasek and Solanumlacteum Vell. All are illustrated, mapped and assessed for conservation status. We also provide a brief analysis of the diversity and endemism of the Geminata clade in Brazil and a key to all 44 Brazilian species.

Descrições detalhadas e lectotipificações são apresentadas para Solanumlacteum Vell. e Solanumapiahyense Witasek. Para todos os táxons são apresentados imagens, mapas e categorias de ameaça. É também apresentada uma breve análise de diversidade e endemismo do clado Geminata no Brasil, além de uma chave dicotômica para as 44 espécies ocorrentes no país.
